A new alternative to produce gibberellic acid by solid state fermentation BABT
Rodrigues,Cristine; Vandenberghe,Luciana Porto de Souza; Teodoro,Juliana; Oss,Juliana Fraron; Pandey,Ashok; Soccol,Carlos Ricardo.
Gibberellic acid (GA3) is an important hormone, which controls plant's growth and development. Solid State Fermentation (SSF) allows the use of agro-industrial residues reducing the production costs. The screening of strains (four of Gibberella fujikuoroi and one of Fusarium moniliforme) and substrates (citric pulp, soy bran, sugarcane bagasse, soy husk, cassava bagasse and coffee husk) and inoculum preparation study were conducted in order to evaluate the best conditions to produce GA3 by SSF. Fermentation assays were carried out in erlenmeyers flasks at 29°C, with initial moisture of 75-80%. Different medium for inoculum production were tested in relation to cells viability and GA3 production by SSF. F. moniliforme LPB 03 and citric pulp were chosen for...

Bioprocessing of some agro-industrial residues for endoglucanase production by the new subsp.; Streptomyces albogriseolus subsp. cellulolyticus strain NEAE-J BJM
El-Naggar,Noura El-Ahmady; Abdelwahed,Nayera A.M.; Saber,Wesam I.A.; Mohamed,Asem A..
The use of low cost agro-industrial residues for the production of industrial enzymes is one of the ways to reduce significantly production costs. Cellulase producing actinomycetes were isolated from soil and decayed agricultural wastes. Among them, a potential culture, strain NEAE-J, was selected and identified on the basis of morphological, cultural, physiological and chemotaxonomic properties, together with 16S rDNA sequence. It is proposed that strain NEAE-J should be included in the species Streptomyces albogriseolus as a representative of a novel sub-species, Streptomyces albogriseolus subsp. cellulolyticus strain NEAE-J and sequencing product was deposited in the GenBank database under accession number JN229412. This organism was tested for its...

Column bioreactor use for optimization of pectinase production in solid substrate cultivation BJM
Linde,Giani Andrea; Magagnin,Glênio; Costa,Jorge Alberto Vieira; Bertolin,Telma Elita; Colauto,Nelson Barros.
This study aimed to determine the influence of process variables and production, of polygalacturonase (PG) and polymetylgalacturonase (PMG) by solid substrate cultivation using a fixed bed column bioreactor. A fractional factorial design (FFD) was used to study the effect of the following variables: microorganism (Aspergillus oryzae and Aspergillus niger), substratum (wheat bran and defatted rice bran), aeration (40 and 60 ml h-1g-1), pectin (5 and 10 g g-1) and nitrogen (urea and ammonium sulfate). Microorganism, aeration and initial pectin were identified in FFD as significant variables (p<0.05) on PG production. A central composed design to optimize PG and PMG productions indicated that Aspergillus niger presents higher PG production; substratum and...

Quality of clonal plants of Piptocarpha angustifolia in different renewable substrates and seasons of the year PAB
Stuepp,Carlos André; Wendling,Ivar; Koehler,Henrique Soares; Zuffellato-Ribas,Katia Christina.
Abstract The objective of this work was to evaluate the effect of different compositions of renewable substrates and seasons of the year on the quality of Piptocarpha angustifolia clonal plants. The experiment was carried out in a completely randomized design in a 4x6 factorial arrangement (four seasons x six substrate compositions). The analyzed substrates were: S1, commercial substrate with pine bark and coconut fiber base (CF); S2, carbonized rice husk (CRH); S3, fibrous and grainy CF; S4, 50% CF and 50% CRH; S5, 30% CF and 70% CRH; and S6, 70% CF and 30% CRH. Minicuttings of 8±1 cm were planted in tubes of 110 cm3 and placed in a greenhouse, during the four seasons of the year. The evaluated variables were: survival and number of roots at 70 days;...

Rice bran as a substrate for proteolytic enzyme production BABT
Sumantha,Alagarsamy; Deepa,Paul; Sandhya,Chandran; Szakacs,George; Soccol,Carlos Ricardo; Pandey,Ashok.
Rice bran was used as the substrate for screening nine strains of Rhizopus sp. for neutral protease production by solid-state fermentation. The best producer, Rhizopus microsporus NRRL 3671, was used for optimizing the process parameters for enzyme production. Fermentation carried out with 44.44 % initial moisture content at a temperature of 30 C for 72 h was found to be the optimum for enzyme secretion by the fermenting organism. While most of the carbon supplements favored enzyme production, addition of casein resulted in a marginal increase in protease yield. Fermentation was then carried out under optimized conditions to obtain the crude extract of the enzyme, which was partially purified by precipitation and dialysis. A 3-fold increase in the enzyme...

Spore production of Beauveria bassiana from agro-industrial residues BABT
Santa,Herta Stutz Dalla; Santa,Osmar Roberto Dalla; Brand,Débora; Vandenberghe,Luciana Porto de Souza; Soccol,Carlos Ricardo.
The purpose of this work was to produce Beauveria bassiana by Solid-State Fermentation using agro-industrial residues and optimizing the cultivation conditions. Refused potatoes, coffee husks and sugar-cane bagasse were tested. The blend of refused potatoes and sugar-cane bagasse (60-40%) with particle size in the range of 0.8-2 mm was used in the fermentation experiments. In Erlenmeyer flasks the best spore production was achieved with the following conditions: incubation temperature 26º C; initial pH 6.0; inoculum concentration 10(7) spores.g-1.dw and initial moisture 75%. In the column type reactor using forced aeration under the optimized conditions, the maximum production (1.07x10(10)spores.g-1.dw) was obtained at the 10th day of fermentation. The...
